CHANGES OF CHROMOSOME NUMBER DURING MEIOSIS IN SACCHAROMYCES CEREVISIAE

Cel

ELABORATION AND COMPARATIVE EVALUATION OF TEST SYSTEMS FOR THE ASSESSMENT OF GENETIC EFFECTS OF ENVIRONMENTAL CHEMICALS AND OF THE PRODUCTS OF THEIR METABOLIC TRANSFORMATION.

THE AIM OF THE PROJECT IS TO FURTHER DEVELOP AND EVALUATE A SYSTEM WHICH ALLOWS THE QUANTITATIVE ESTIMATION OF ANEUPLOIDY (FOR CHROMOSOME V) AND GAMETE DIPLOIDISATION IN SCCHAROMYCES CEREVISIAE DURING MEIOSIS WHICH HAS BEEN ALREADY ANALYSED FOR THE SPONTANEOUS OCCURENCE OF SUCH EVENTS.

IT PLANNED TO INVESTIGATE THE EFFECT OF CHEMICAL AGENTS ON THE INDUCTION OF NUMERICAL CHROMOSOME CHANGES WITH PARTICULAR REGARD TO DNA LIGANDS, CHEMICALS AFFECTING MEMBRANES AND/OR THE SPINDLE APPARATUS, AND NON-MUTAGENS ENDOWED WITH POTENTIAL SPECIFIC EFFECTS ONLY ON CHROMOSOME SEPARATION DURING MEIOSIS.

IN PARTICULAR, THE FOLLOWING CATAGORIES OF CHEMICALS WILL BE STUDIED:

1) COMPOUNDS SELECTED ACCORDING TO THEIR MECHANISMS OF ACTION

THE AIM OF THIS APPROACH IS NOT ONLY TO DISCOVER NEW NONDISJUNCTANTS BUT ALSO TO BETTER UNDERSTAND THE BASIC MECHANISMS CAUSING THE FAILURE OF CHROMOSOME CENTROMERES TO SEPARATE AT MEIOSIS.

THE COMPOUND WILL BE GROUPED ACCORDING TO THEIR MODE OF ACTION AND SPECIFICITY OF TARGET (AGENTS BINDING TO DNA, MEMBRANE-AFFECTING COMPOUNDS, AGENTS INTERACTING WITH THE SPINDLE FIBRES).

2) NON MUTAGENS CAUSING NON-DISJUNCTION

IT SEEMS RELEVANT TO FIND SOME COMPOUNDS THE ONLY EFFECT OF WHICH IS NONDISJUNCTION AND/OR GAMETE DIPLOIDISATION, AS SUCH CHEMICALS WILL ESCAPE THE USUAL ENVIRONMENTAL MUTAGENIC SCREENING (TPA, ETC.).

3) COMPOUNDS SUSPECTED TO BE LINKED WITH A REDUCTION OF FERTILITY

SOME COMPOUNDS SUSPECTED FROM EPIDEMIOLOGICAL DATA TO CAUSE UNFERTILITY OR TO REDUCE FERTILITY BE TESTED, SUCH AS HALOTHANE.
