Objective EVALUATION OF RISKS FROM RADIATION-INDUCED CANCER. DETERMINATION OF THE ROLE OF DNA METHYLATION. IT IS PROPOSED TO INVESTIGATE WHETHER AND HOW IN AN IN VITRO CELLULAR SYSTEM, CAPABLE OF CELL DIFFERENTIATION UNDER CONTROLLED CONDITIONS, EXPOSURE TO IONIZING RADIATION MODIFIES, IN A CORRELATED MANNER, THE DNA METHYLATION PATTERN AND THE CULTURAL AND/OR DIFFERENTIATION CHARACTERISTICS. Research work includes: studying the effects of sublethal exposure to X-irradiation on specific gene expression by cultured cells endowed with a deoxyribonucleic acid (DNA) methylation sensitive tendency to undergo terminal differentation; studying the effects of DNA methylation inhibitors on the susceptibility to radiation damage of cultured mammalian cells; evaluating single strand breaks occurring in nuclear DNA during cell differentiation and/or exposure to X-rays; identifying factors which regulate, within chromatic structure and at the single gene level, the susceptibility to in vitro methylation of particular CpG sequences in DNA.A STUDY OF THE LEVEL OF METHYLATION OF DNA IN THE REPAIR OF RADIATION INDUCED DAMAGE, ITS EFFECT ON DIFFERENTIATION, TRANSMISSION TO DAUGHTER CELLS AND ITS POTENTIAL ROLE IN CELL TRANSFORMATION.
