Development and investigation of systems for the quantification of radiation induced carcinogenesis in humans

Objetivo



In the last few years there has been a growing realisation that studies of initial events in carcinogenesis must include consideration of apoptosis or programmed cell death. This process is under the control of cmyc and p53 genes, among others, and these genes are also highly invoived in carcinogenesis. The aim of this additional proposal which is from a world class laboratory in the Apoptosis field, is to assess the importance of death by apoptosis in human cultures produced and irradiated by other members of the group. Since Apoptosis is thought to be an "altruistic suicide" of radiation damaged and mutated cells, it is clearly important to assess its prevalence in any study of initiating events in carcinogenesis. Apoptotic bodies are common in cultures seen by other members of the group and correlate with expression of cmyc and p53 genes. Delayed apoptosis is also a known late effect of irradiation. This proposal, if funded, will quantify the extent of apoptosis in a variety of human cell systems with respect to dose, radiation quality and time. The methodological approaches to be used include quantitative analysis of the status of nuclear DNA. This is of crucial importance in defining the dose response relationship. The laboratory will also use two of its novel methods developed for quantifying levels of apoptosis - response to detergent treatment and selective staining by acridine orange. The contribution of the Prague Laboratory will provide another dimension to the quantification of carcinogenic initiation, ie., the integrity of the programmed cell death system in control and irradiated cells. This system if functioning correctly, should remove potentially transformed cells from the survivor population.
