Assessment of human exposure to urban environmental carcinogenic pollutants

The participation of Dr. Sram in this research programme was originally included in the submitted proposal (PL910591), and full details of his intended contribution are included therein. For technical reasons Dr. Sram could not be funded in the programme's contract (EV5V-CT92-0198). This application is therefore for funds to allow the incorporation of Dr. Sram into the ongoing programme.
The basis of the programme is the development of novel biomonitoring approaches for carcinogens, which will be applied to samples from populations exposed to environmental pollutants. Dr. Sram's proposed contribution (which was considered of essential importance by the other participants) involved three areas. Firstly, he plans to develop new methods, involving the combination of HPLC and 32p-postlabelling, for the detection of oxidative damage in DNA. The second contribution of Dr. Sram will be in the improvement and application of methods for detecting by 32P-postlabelling the damage caused by carcinogenic polycyclic aromatic hydrocarbons with DNA, which will be applied to DNA from various human tissues. Dr. sram additionally intends to supply biological samples from a population exposed to environmental pollution in N.Bohemia and from a population from a less polluted area in S.Bohemia. Blood samples will be collected from mothers and from newborns (cord blood), and placenta samples will also be made available to the other participants for postlabelling and mass spectral analysis of DNA damage. Results will be compared with those from samples collected by participants 2 (DK), 3 (GB) and 4 (GR). Dr. Sram's contribution is considered of considerable relevance towards the aims of the programme:(i) design of novel biomonitoring methods, (ii) comparison of carcinogen-induced damage to DNA in different human tissues and (iii) establishment of baseline data for such damage in control human populations. As the involvement of Dr. Sram was envisaged at the time of design of the research plans of the other 4 participants inthe programme, no extra funding is required by these participants. This request for resources is therefore solely to enable Dr. sram to carry out his proposed participation in the programme.
