Development of Methods for the Clonal Propagation of Elite, Disease Resistant Palms by Somatic Embryogenesis

Obiettivo



Overcome present problems in the culture of coconut tissues to improve somatic embryogenesis and to ensure mass production of plantlets.
1)Identify embryogenic potential using cellular and molecular markers to obtain highly regenerative cell lines.
2) Study growth substance control of somatic embryogenesis from initiation to embryomaturation
3) Study vitroplant physiology and optimal conditions for plantlet development and transfer to ex vitro environment.
4) Implement technological advances through exchange of information and personnel. Provide training facilities, particularly for partners in tha Philippines, Mexico and other coconut growing countries.
a) conventional coconut breeding programmes can only produce disease-tolerant and high-yielding stock very slowly. Coconut growing countries wish to propagate locally-adapted palms which have desirable characteristics for their particular environment and market. A widely applicable method of clonal propagation such as somatic embryogenesis woukd enable such countries to produce high quality populations to renew their ageing plantations.
b) such a system is not available despite many years of work. It is now proposed link the log experience gained in tissue culture of coconut at wye and ORSTOM/CIRAD with that of Hannover University to exploit the new techniques which are being successfully used with other crops. CICY in Mexico and Albay Research Center of the PCA in the Philippines will be involved in the project. c) the project will focus on the marker-assisted development of protocols to establish highly embryogenic cultures on both solid and liquid media. Techniques of histology and growth regulator analysis, using tracers, will be applied to follow and better manage somatic embryo induction and development. d) DNA markers will be developed and used to confirm the clonal fidelity of the regenerants (ramets).
e) particular attention will be paid to the later stages of embryo development in order to produce normal plantlets, and also to their successful establishment ex vitro.
f) the new technology arising from this project will be shared with all the partners not only routinely through regular reports and visits but also at the workshops which are planned during the course of the collaboration. These are of major importance in achieving the full benefits of the program.
