Manipulation of Oligosccharide Associated Inflammatory Mechanism to Treat Arthritis

Obiettivo



Chronic arthritis is of major public health importance to the European Community and this Concerted Action brings together laboratories to form a cohesive unit that will uniquely be able to carry out the research proposals described to tackle arthritis.

The European laboratory that has been formed by this Concerted Action focuses upon the four areas of glycobiology.

Glycoproteins (Immunoglobulin G and acute phase proteins)
Oligosaccharide molecular modelling. Cell adhesion molecules. The glycosyltransferase This proposal seeks to expand and consequently make more scientifically effective our recently established Concerted Action involving oligosaccharides and arthritis. The areas selected for expansion fall into two crucial areas:

Oligosaccharides and Proteins.

(a) The role the oligo-saccharide moiety of human IgG-Fc plays in determining i ternary structure will be elucidated together with insight into its interaction sites for ligands.
(b) The glycosylation dependent biological functions of rheumatoid factors will be determined.
(c) Characterisation of IgG glycosylation in psoriatic arthritis will be determined.

The Glycosyltransferases.

(a) A system for modulation of protein oligosaccharide characteristics by glycosyltransferase gene transfer will be developed, together with clinical protocols for gene therapy.
(b) Modulation of the expression of the a-1,3/4-fucosyltransferase (FT III) responsible for the synthesis of Lex and Lea structures will be investigated.

These areas have been selected for expansion as they integrate well into the main focus of the Concerted Action, which is to elucidate the mechanisms wh inflammatory process and, how these mechanisms may be inhibited to bring about amelioration of the inflammation process resulting in arthritis.
