Exposure to genotoxic chemicals in the steel industry: detection of groups at risk

The aim of the study is to assess the usefulness of combining tests for exposure to genotoxic chemicals with the measurement of a series of effect markers for detecting groups of workers who may suffer excessive exposure to genotoxic chemicals. The aim of the statistical analysis is to examine whether the prevalence of positive markers differs between the two groups (exposed and control) and whether these effects can be related to the intensity of exposure.

Groups of works who might be exposed to genotoxic chemicals, together with control groups (about 600 subjects in all) will be identified in various steel works. Various factors which are known to influence the background level of genotoxic risk will be assessed by means of a detailed questionnaire.

The markers to be monitored can be classified in two groups:

A.Markers of exposure (specific or aspecific).
B.Markers of effects (biological parameters in the blood and urine).

METHODS AND MEANS BY WHICH THE AIMS ARE TO BE ACHIEVED

The questionnaire designed to detect the various factors known to influence the background level of genotoxic risk will concentrate in particular on smoking habits, medicament and alcohol consumption, nutritional factors, and the use of respirators.

Specific markers of exposure will be assessed through air analysis (static and personal sampling) by high pressure liquid chromotography, and through biological monitoring; aspecific markers to be studied will be assessed by means of urine samples (mutagenicity and thioethers).

Markers of effects to be monitored will be pseudouridine in urine, carcinoembryonic antigen in serum, a-foetoprotein in serum, tissue polypeptide antigen in serum, and other less specific markers.
