Measurement of lipid peroxidation during respiratory inflammation in coal workers

Objetivo

The subject of this research is the role of lipid peroxidation (an intra-cellular oxidative biochemical reaction) in respiratory inflammation in coal workers. The aim is to determine whether their lipid peroxidation metabolites have increased, whether they reflect recent exposure, whether they are influenced by cigarette smoke and whether they could be used in medical surveillance of exposed workers.

The study will cover three groups:

- miners not suffering from pneumoconiosis who are currently exposed to coal dust (smokers and non-smokers);
- workers not exposed to dust or other pulmonary irritants (smokers and non-smokers);
- active or retired miners suffering from pneumoconiosis (smokers and non-smokers).

There will be thirty subjects in each group.

In principle, there will be one sample (air and urine) taken from each worker.

The results will be analysed using the "Statistical Package for Social Science" under the supervision of a statistician.

METHODS AND MEANS BY WHICH THE AIMS ARE TO BE ACHIEVED

Exhaled air will be analysed using the original method developed at IUMHT in Lausanne.

Briefly: after double concentration of the air sample by cold capture, the light hydrocarbons (including pentane) will be analysed by gas-phase capillary chromatography with detection by flame ionisation.

Malondialdehyde in the urine will be analysed by HPLC (high performance liquid chromatography) in accordance with the method adapted by Knight.

A quality control programme will be set up to ensure the reliability of the results.
