Objective To collect in Europe rheumatoid arthritis (RA) families : - with affected sib-pairs to map RA susceptibility loci - with RA cases and both parents for linkage disequilibrium mapping of the genes involved.Rheumatoid arthritis (RA) is a chronic arthritis with a prevalence of 0.5% in adults. There is a genetic predisposition to this multifactorial disease. Association with HLA class II antigens accounts for a third of it. The aim of the project is to localize the other genes involved, taking advantage of the new dense map of the human genome. Thanks to the collaboration between 7 European states, 330 affected sib-pair families and 230 families with RA cases and both parents will be collected. The GENETHON facilities will be used for genotyping 400 evenly spaced highly polymorphic markers in affected sib-pair families. The genotype analysis will allow susceptibility loci, showing an excess of allele sharing by affected sib-pairs, to be roughly localized. Association studies with markers of these loci, using the families with both parents, will then be performed for linkage disequilibrium mapping of the genes involved. The understanding of the genetic predisposition to RA should allow a breakthrough in the understanding of the pathophysiology of this crippling disease, leading to a therapeutic improvement.
