The role of ectomycorrhizae in stress tolerance of forest trees

The main objectives are to study various aspects of tree root and mycorrhizal interactions, in order to provide a firm data base on the role of ectomycorrhizae in stress tolerance of forest trees. This will then be used for estimating the impact of adverse soil conditions on trees.

The project will investigate the influence of stress in rhizosphere, on rhizosphere microflora, ectomycorrhizae and tree seedlings. In addition to basic information about the role of ectomycorrhizae in nutrient and water acquisition, mechanisms by which mycorrhizal seedlings may resist adverse soil conditions will be investigated. The project will include investigations of the uptake and localization of mycorrhizal tree seedlings. The effects of toxic metals on the uptake of mineral nutrients will also be covered. In order to contribute to the understanding of the principles by which ectomycorrhizae may increase the stress tolerance and its ecological relevance, these studies will use experimental parameters which reflect the conditions of natural forest sites.

Experiments will be carried out using Norway spruce seed of the same origin and Paxillus involutus as common symbiots. In 2 subprojects, other tree species will also be used. In several experiments in addition to Paxillus involutus, other fungi will be included. The work will be carried out under controlled environmental conditions, especially for the roots. The techniques used to cultivate the symbionts will include pure cultures, nutrient solutions cultures with various solid media (sand, perlite) and soils (natural and artificial). The content of nutrient, heavy metals and aluminium in mycorrhizal fungi and plant tissues will be estimated using atomic absorption spectometry, isotopic efflux analysis, X-ray microanalysis (energy dispersive X-ray analysis (EDX) and nuclear magnetic resonance techniques).
