Deposition and study of structure and functional properties of Langmuir-Blodgett films of light-sensitive proteins

Objectif



This project will be concerned with the study of the behaviour of the protein monolayers at the air/water interface; determination of the conditions for their transfer onto solid substrates; and investigation of their structure and function at the solid substrate. The work will be performed using bacteriorhodopsin and photosynthetic reaction centres from different bacteria, such as Rhodobacter sphaeroides, Rhodopseudomonas viridis and Chromatium minutissimum. p-A isotherms, surface potential measurements and ellipsometry will be applied for the investigation of protein monolayers at the air/water interface.

The quality of the deposition and its reproducibility will be estimated by nanogravimetric measurements. The X-ray diffraction and STM study will be used for the characterisation of the structure of the deposited films. CD measurement will enable the changes in the secondary structure of the protein in the film to be estimated. Optical spectroscopy and surface potential measurements will be applied for the study of the activity of the proteins in LB films. Thermal stability of the proteins in LB films will also be investigated.
