Use of physical gene mapping to study genome organisation and intercommunication in natural and transgenic Nicotiana tabacum

The project aims to bring together the large volume of cytogenetic and molecular data available for Nicotiana tabacum by mapping known genes and DNA sequences using in situ hybridization to identify marker chromosomes in the N. tabacum karyotype. This will provide a bridge between cytogenetic and molecular approaches, without which the considerable potential of N. tabacum as a research tool cannot be maximized. A major application of this research for plant genetic engineering will be the provision of reference points to allow monitoring of alien DNA introduced into N. tabacum by direct gene transfer or somatic hybridization.

Physical markers were already identified for the two component genomes of N. tabacum and points of recombination between them. Techniques will be introduced to map genes and DNA sequences of low copy number, both simultaneously and sequentially, using a combination of differently labelled DNA probes. Physical markers will be used to investigate the nuclear domains of genes, transposable elements and repeated DNA sequences in relation to ancestral genomes and sites of intergenomic recombination. The results will be used to examine chromosome structure and reorganization in sexual somatic hybrids, and to explore the role played by transposons in genome modification and intergenomic communication in hybrids.

An important feature of the project will be to transfer the technology of non-radioactive in situ hybridization to the Ukrainian group. This will enhance work concerned with detection of alien DNA and genom reorganization in genetically engineered plants.
