Biomonitoring of occupationally exposed populations to acrylonitrile

Foreseen Results

The applied scientific goal of the proposed research is to help in identifying on a cost-benefit basis the most accurate and practical methods to assess hazardous occupational exposures.
The present proposal is aimed to evaluate the genetics effects of acrylonitrile in two occupational exposed populations from two plants (one in Portugal and one in Czech Republic). It involves 6 laboratories, 3 from EU-member states (Portugal, Belgium and United Kingdom) and 3 from Central Europe countries (Czech Republic, Slovakia and Hungary). Besides aiming the contribute to a better understanding of the hazardous exposure of man to acrylonitrile, it aims to help validating and optimizing the use of a combination of methods applied to human populations exposed to genotoxic compounds. Three main compartments related to the dose or effects of the hazardous compounds will be evaluated using various assessment methods :

Internal dose : mutagenicity in urine, indicators of oxidation stress, induction of cytochrome P450;
Biological effective dose : haemoglobin adduct, DNA excision products;
Early biological effects : chromosomal aberrations, sister chromatid exchanges, micronuclei and DNA damage (Comet assay).
