Radiation effects and their treatment on the connective and vascular tissues in various organs

Substantial progress have been made in the clinical description, predisposing factors and pathological features of RT-induced late cognitive dysfunction after conventional cerebral RT. For the first time, an experimental model of this syndrome has been developed which should be useful to understand its pathogenesis.
An acute model of cognitive dysfunction after whole body irradiation has been developed which seems quite useful to test radioprotective agents. In the future we hope to try these radioprotective agents in the chronic model.
THE RESEARCH PROJECT CONCERNS I)THE STUDY OF PATHOLOGICAL, PHYSIOLOGICAL, AND BIOCHEMICAL PARAMETERS, ON CLINICAL OR EXPERIMENTAL MATERIAL, IN ORDER TO DEFINE THE EVOLUTIVE POTENTIAL OF RADIATION INDUCED DAMAGE TO THE VASCULAR AND CONNECTIVE OR SUPPORTIVE TISSUES TISSUES IN VARIOUS ORGANS, II) THE DEVELOPMENT OF PARACLINICAL TESTS FOR DIAGENOSIS AND THE EVALUATION OF SEVERAL PHARMACOLOGICAL APPROACHES TO PREVENT AND/OR TREAT THESE DAMAGES AFTER ACCIDENTAL IRRADIATION. THE ORGANS CONSIDERED WILL BE THE LUNG (PARTIAL BODY), LENS, OSSEOUS AND TENDINOUS TISSUES, MUSCLES, VASCULAR TISSUE AND GLIAL CELLS IN THE CENTRAL NERVOUS SYSTEM (CNS). THE BIOLOGICAL AND EXPERIMENTAL APPROACH WILL FOCUS ON THE CHARACTERIZATION OF PARACRINE REGULATIONS BETWEEN MESENCHYMATOUS AND EPITHELIAL CELLS (CHEMICAL MEDIATORS, HORMONES, CYTOKINES AND GROWTH FACTORS) AND ON MECHANISMS OF TISSUE POROTECTION BY SEVERAL COMPOUNDS. THE THERAPEUTIC APPROACH ENVISAGES THE LOCAL ADMINISTRATION OF PHARMACOLOGICAL COMPOUNDS CAPABLE TO PREVENT OR TO TREAT THE CONSEQUENCES OF ACUTE OVEREXPOSURES TO IONIZING RADIATIONS IN ACCIDENTAL SITUATIONS. VARIOUS MODALITIES OF ADMINISTRATION OF COMPOUNDS KNOWN TO REVERSE THE POST IRRADIATION FIBROTIC EVOLUTION, SUCH AS SUPEROXYDE DISMUTASE (SOD), WILL BE DEVELOPED ACCORDING TO THE TARGET ORGANS: AEROSOLS FOR FOR LUNG FIBROSIS, TRANS CUTANEOUS INFUSION FOR DEEP LESIONS, ETC...) AND EVALUATED WITH OBJECTIVE CLINICAL AND PARACLINICAL INDICES. THE ACHIEVEMENTS EXPECTED FROM THIS PROJECT ARE I) A BETTER ASSESSMENT OF THE EVOLUTIVE POTENTIAL OF RADIATION INDUCED FIBROSIS, SCLEROSIS, THROMBOSIS AND NECROSIS AND ASSOCIATED PATHOGENESIS (PHYSIOPATHOLOGY, DIAGNOSIS, PROGNOSIS) AND II) AN EVALUATION OF DIFFERENT TREATMENTS TO PREVENT, REVERSE OR LIMIT THE PROGRESSION OF VARIOUS PROCESSES.
