Evolving hyper-recombinase low-fidelity reca

Objectif

I enthusiastically recommend Mr. Ariel B. Lindner (Ari) to the EC Marie Curie Individual Fellowship, to support his postdoctoral studies in Prof. Miroslav Radman in Necker, Paris. I supervised Mr. Lindner in his MSc. and PhD studies, the duties towards which he has just accomplished at the Department of Immunology The Weizmann Institute of Science. Ari is one of the very few outstanding students I ever had or knew. He is extremely intelligent, creative, equipped with an excellent knowledge-base and performs very well at the bench. Educated as Chemist, he has a sound knowledge and understanding of the basic components and rules that govern our solid world. Driven to know more, he turned into biology and joined our basic research endeavors, trying to utilize the huge antibody repertoire to generate new type of catalysts such as catalytic antibodies and understand their mechanism of action. His MSc thesis was innovative and original and granted him one of the best award of our graduate school. In his PhD studies, in addition to the characterization of mode of action of esterolytic antibodies, he focused on the generation and selection of catalytic antibodies out of large libraries. Along this line, he adopted the new technology of Molecular Evolution developed by Dr Dan Tawfik at the MRC Cambridge. This powerful technology allows the selection of new catalysts according to their function. Ari has already developed a model system, using catalytic antibodies we have established before, and I hope that in the period left before his departure for his post doctorate in Paris, we shall be able to submit these original and innovative accomplishments for publication. Ari's PhD has been evaluated again as extremely innovative. His achievement, published in leading journals received excellent reviews and responses. Using kinetic, structural and molecular-biology approaches he discovered a unique way certain antibodies employ to catalyse their Substrate/Moreover, Ari succeeded to pin-point it to certain residues in the antibody binding site. The multi-facet nature of his study in this inter-disciplinary project, made him an expert in various fields and technologies such as chemistry, enzymology, chemical immunology and molecular biology. These, together with his intelligence, originality and creativity makes him unique amongst his peers. Ari performed part of his studies independently of me, as reflected in his publications, He has been invited to work in a few of the leading institutions (such as MRC, Cambridge with Prof. Alan Fersht, Scripps Clinics, La Jolla with Prof. Richard Lerner). For his post doctorate research, Ari has been accepted to work in the group of Prof. Miroslav Radman in Necker, Paris. Miroslav Radman is a leading geneticist who pioneered the notion of the SOS stress-induced response; his current seminal contribution to the understanding of mismatch repair and recombination mechanisms and their implication in disease (as cancer) and evolution has lead Ari to choose this place for his training. The research he will be doing there involves with the genetic evolution in lower organisms, a field where the molecular mechanism is probably simpler to dissect, yet is certainly related to the broader aspect of Mr. Lindner interest which is developmental evolution. Based on my recognition of his outstanding performance and merits, I am...
