Evaluation of the role of plant cell wall degrading enzymes on yield and quality of oil produced by cold pressing of low-oil-producing crops

Objective

The primary objective of this project is to develop an efficient and novel enzymatic method to extract nutritionally valuable oil from sorrel, pomegranate and tomato seeds by cold pressing, without losing its nutritional properties. To achieve this objective, we aim to
(1) identify and characterize the carbohydrate components of sorrel, pomegranate and tomato seed shells;
(2) determine the effect of cell wall degrading enzymes on carbohydrate part of the seed shells;
(3) optimise the conditions for the enzymatic hydrolysis of carbohydrate part of the seed shells;
(4) select ideal conditions for the extraction of oil from enzymatically processed seeds, and;
(5) to evaluate the composition, yield and nutritional value of oil extracted from sorrel, pomegranate and tomato seeds with and without enzymes under cold pressing conditions.

This proposal also aims to integrate scientific and technical expertise from INTAS and NIS countries in order to exchange the information and technology in a most effective manner.
The consortium of this joint research proposal includes two research contractors from INTAS and three research proposers from NIS countries. The multidisciplinary approach of this proposal brings expertise from enzymology, protein chemistry, plant cell wall biochemistry, physical chemistry, human nutrition, food technology and biotechnology. All research partners have considerable knowledge and experience in their field of research and will contribute effectively for the successful completion of this project. The collaboration will also facilitate the visit of young scientists from NIS countries to INTAS partners' laboratories to carry out research and obtain the state of art training.
During the initial stages of this project, the carbohydrate composition of sorrel, pomegranate and tomato seed shells, their relation and the capability of swelling under different process conditions will be defined. Cellulose, hemicellulose, lignin and tannin of these seed shells will be extracted and characterized. The effect of cellulases and hemicellulases on original seeds of sorrel, pomegranate and tomato will be studied under different conditions (time, temperature, pH, enzymes concentration). Optimal conditions for enzymatic hydrolysis will be determined. Conditions for the extraction of high quality oil with maximum yield will be defined. Also, the optimal temperatures and humidity of the material under the cold pressing conditions will be defined. The optimal sizes and conditions for seed grinding will be established together with particle dispersity in order to define the greater effect of enzymes at different concentrations of ferment and substrate. The comparative analysis of composition and yield of sorrel, pomegranate and tomato oil will be carried out on pre- and post- the fermentative processing under optimal conditions. The biological and nutritional values of oil seed products will be detected after their fermentative and cold pressing processing. The assimilability of these products by humans and the effect of protein-inhibitors on their composition will be predicted and their inhibitor activity in relation to trypsinum and chymotrypsinum will be defined.
The development of a novel oil extraction technology using enzymes under cold pressing conditions will enable us to increase the production of food products having valuable biological and nutritional properties in addition to the rapid assimilation of these products by humans.
