Properties and kinetics of adhesion of myelin basic protein isoforms

Objectif

The process of demyelination accompanies many neurological diseases. The intensity of demyelination and induction of inflammation depends on the adhesive ability of myelin basic protein (MBP) to oligodendrocytes and T-lymphocytes. The proposed project is aimed at studying the influence of various types of post-translational modifications (phosphorylation, amination, ADP-ribosylation) on the adhesive properties of MBP. The project is directed towards the following points:
(i) purification and preparation of various modified (phosphorylated, ADP-ribosylated, deaminated) forms of MBP;
(ii) Study of kinetics of adhesion of different modificated isoforms of MBP with artificial membranes and with lymphocytes from normal and multiple sclerosis (MS) patients;
(iii) Investigation of secretion of cytokines from lymphocytes of healthy and MS patients under the action of the various isoforms of MBP. Results will hopefully reveal the functionally active form of MBP, which participates in the process of myelin repair, and also the immunogenic form of MBP, which causes restriction of T-lymphocytes.
