Boron compounds for radiodiagnostics and radiotherapy

This project deals with the exploration of functionalised boron cage compounds for use in radiodiagnostics and radiotherapy. Various derivatives of such compounds are here proposed as in vivo carriers of radionuclides. Especially compounds with stable bonds between the radionuclide and the boron cage, and with functional groups, which allow conjugation to biomolecules, will be studied.

Together the 7 teams involved will specifically explore the following three tasks:
1.The use of closo-dodecaborate anion [B12H12]2- as linker for radiohalogenation of macromolecules. Different functional derivatives of the closo-dodecaborate anion will be synthesized and their halogenation with various radiohalogens (bromine, iodine, astatine) will be studied;
2.Radioimmunoconjugates with proteins, antibodies, shorter peptides containing radiohalogen-bearing closo-dodecaborate cage will be prepared using indirect and direct approaches. The selection of radiohalogen label will be made in accordance with the objectives of individual biochemical experiments and the "time window" required;
3. Metallacarborane complexes of different types will be used as carriers of various radioactive metal isotopes. The complexes which will be prepared include the dicarbaundecaborate complexes of cobalt, iron, nickel, and copper containing functional groups for conjugation with macromolecules, tris(2-mercapto-1,2-carboran-1-ylmethyl)amine gallium, and copper(I) bis(1,2-carboranyl diphosphine) complexes. Radioconjugates of the functional derivatives of metallabis (dicarbollides) with macromolecules will be prepared. Stability and immunoreactivity of the radioimmunoconjugates prepared will be studied.
