Objetivo The study proposed by 3 Russian, 2 French and 1 German teams will be devoted to evaluation of short-duration intermittent +2Gz centrifugations as countermeasures to prevent the development of some microgravity effects in rat brain and spinal cord arising during head-down tilt as result of the lack of support load on hindlimbs and liquid redistribution to anterior part of body. The aim of the study will be to investigate the effects of intermittent centrifugations aboard the Space Station as countermeasure decreasing the microgravity influence in man in space. Rats will be exposed to a 24-day head-down tilt with a 1-hour +2Gz influence, administered daily or every two days, this countermeasure beginning from the 6th to the 24th day of the experiment. The hindlimb area of brain somatosensory cortex and spinal cord anterior horn at level L1-L3 will be studied as structures receiving hindlimb proprioceptive input. The cerebellar nodulus and medial vestibular nucleus will be studied as structures receiving afferent vestibular input. In the brain the choroid plexus, raphe dorsalis nucleus, locus coeruleus, supraoptic nucleus, median eminence and neurohypophysis as structures connected with cerebrospinal fluid (CSF) production and regulation will be investigated, also as ependyma and arachnoid Pacchinian granulations involved in CSF circulation and CSF escape in superior sagittal sinus.For estimation of blood vessel state in the brain at prolonged redistribution of body liquor induced by head-down tilt and at the facilitation of blood outflow from brain induced by intermittent +2Gz centrifugation the meninges, superior sagittal sinus and blood vessels of endbrain, middle brain, medulla oblongata and cerebellar hemispheres will be studied. Hormones content in blood, aquaporins in kidney and mass of thymus adrenals and spleen will be parallelly studied. Investigations will be performed using immunocytochemical, in situ hybridisation, radioimmunological, electron microscopic quantitative cytochemical, morphometrical and light microscopical methods.The realization of the proposed study will give the oppotunity:- to elucidate mechanisms of carrying out and regulating cerebrospinal fluid secretion, circulation and escape in rat brain under1) prolonged reduction of blood outflow from brain induced by head-down tilt;2) facilitation of blood outflow from brain induced by intermittent +2Gz centrifugation;- to elucidate mechanisms of adaptation in spinal cord, somatosensory cortex and vestibular structures of rat brain to1) prolonged decrease in hindlimb proprioceptive input induced by head-down tilt;2) increase in hindlimb proprioceptive input induced by intermittent +2Gz centrifugation;- to estimate the adaptative response of the blood vessel system in rat brain under1) prolonged difficulty of blood outflow from brain induced by head-down tilt;2) facilitation of blood outflow from brain induced by intermittent +2Gz centrifugation;- to use intermittent and short +2Gz centrifugations as a countermeasure for preventing or reducing head-down tilt effects on rats ;- to obtain data for the development of intermittent +Gz centrifugations algorithm allowing an application for artificial gravity creation by means of centrifuge rotation aboard the International space station.
