Objetivo The aim of this study is the construction of a model of the immunocomplex between the MIR decapeptide and anti-AChR autoantibodies So that these data may be used for the design of high affinity therapeutic antibody fragments. The known NMR structure of the MIR decapeptide, when bound to the antibody molecule, will be used for docking experiments to the known crystal structures of Fab198 and Fab192. The crystal structures of Fab192 and Fab198 subsequently wiII be used in conjunction with the molecular modeling structures of two other anti-AChR monoclonal antibodies (Fab35 and Fab195), in order to combine elements of their structure with known characteristics of their function, The above experiments will show the nature of the interaction between the monoclonal antibodies and the antigen (AChR) and important residues for this reaction wiII be revealed. 3.
