Objectif The subject of this study is mRNA expression profiling of differentiating murine hippocampal neurons. A cDNA microarray approach will be used to simultaneously quantify expression levels of about seven thousand of mRNAs at morphologically and functionally defined stages of neuronal differentiation. A culture system will be used in which neurons undergo differentiation, in a well-coordinated manner, to a fully polarised pyramidal phenotype with functional synapses. The objectives of this study are:1. quantitative analysis of expression of a large mRNA set at defined stages of neuronal differentiation2. Identification of genes that are regulated in the differentiation process3. Cluster analysis of genes according to their expression patterns. The results of this project will form a basis for future work; on regulatory mechanisms underlying coordinated expression of genes in differentiating neurons; and on functional relationships of protein products of the developmentally regulated genes.
