HIGH PERFORMANCE BIOSEPARATION METHODS FOR PROTEOMICS BASED BIOMARKER DISCOVERY

Objective

Interdisciplinary science and technologies have converged in the last few years to create exciting challenges and opportunities involving novel, integrated separation systems to facilitate comprehensive analysis of complex biological samples. Bioseparation science is an essential toolset that enables entering from the genomics era to the proteomics field. The aim of the proposed research and training project is to develop and implement high performance bioanalytical techniques for the field of proteomics based biomarker discovery using single- and multi-dimensional separation methods. One of the main focuses is the establishment of novel column technologies for differential migration processes in micro-channel based bioanalytical systems, such as capillary electrophoresis, capillary electrochromatography and microfabricated separation devices, respectively connected to mass spectrometry. Identification of quantitative and/or qualitative protein expression differences as well as characterisation of various cell proteomes will significantly advance biomedical research and diagnostics. Our novel, high performance bioseparation approaches will be applied to reveal global changes in gene expression at the proteome level. Analysis throughput will be increased by multiplexing and system integration provided by contemporary microfabrication approaches. Up and down-regulation, as well as appearance and disappearance of specific proteins or groups of proteins encountered in cellular systems subjected to changes in such physiological conditions as cancer will be individually screened and identified in order to find surrogate biological markers to be used in early disease diagnostics and speed up clinical trials. This Marie Curie Chair award would practically support the establishment of a new bioseparation science research and training centre in the Central European region for outstanding young researchers from the European Community including the newly joined EU countries.
