UNRAVELING CIRCADIAN ACTIVATION LOOP IN CHROMATIN REMODELLING

Obiettivo

The major goal of this proposal is the improvement of the scientific understanding of the molecular mechanisms that control the mammalian circadian clock. The circadian clock has an impact on several aspects of mammalian physiology of and work in this field has additional implication at the social level. Workers with night shifts, travellers experiencing jet lag, sleep disorders and finally the more and more evident role that circadian time has for an effective cancer therapy, indicate that this proposal has an added value for the community. This project will be realized through the three following objectives, which will explore novel and original aspects of the clock field:
1) To investigate, if circadian control occurs at the chromatin level;
2) Identification of chromatin remodelling factors involved in activation and repression of clock genes;
3) Study of the role of metabolite intermediates in the activation of clock gene transcription. These objectives will be achieved by using well-established methods spanning from molecular biology and biochemistry to the use of animal models. The scientific relevance and quality of the objectives is remarkable: they will open new perspectives not only towards the understanding of the mechanisms by which the molecular clock operates, but also towards potential therapeutic approaches for sleep disorders, seasonal depression, and cancertherapy. The second goal of the proposal is to offer to the applicant top-level post-doctoral research training and introduce him to the circadian clock field. The host institution, the IGBMC, provides a superb training environment in molecular, cellular and developmental biology for young researchers. The host laboratory of Dr. Sassone-Corsi has an international prominent position in the field of circadian biology. The applicant will benefit tremendously from this training: he will complement his competences and his interpersonal skills at the professional level.
