HIGH-THROUGHPUT STRUCTURAL STUDIES OF PROKARYOTIC ACTIVE SECONDARY TRANSPORTERS

The proposed project is aiming at solving the crystallographic structure of some of the prokaryotic membrane proteins belonging to the family of Active Secondary Transporters, an important and heterogeneous class of symport/antiport systems involved in cel l homeostasis and conserved across all reigns of living organisms. Given the difficulty encountered in membrane protein production and crystallization, a High-Throughput approach will be used, in order to screen the highest possible number of different exp ression, solubilization and crystallization conditions and to enhance the probability of success for a few targets out of a starting set of about 100 cloned genes. If determining even only one structure would be an important scientific result, given the ve ry low number of membrane protein structures deposited in the Protein Data Bank, obtaining a few of them would result in a significant improvement in understanding the structural, and hence functional, differences between members of this protein family. Fu rther on, this project will lead to the optimisation of High-Throughput techniques towards membrane protein production and crystallization and will provide a large amount of data to be used for rationalizing the parameters that dictate successes and failur es in membrane protein biochemistry.
