Time-resolved diffusive X-ray scattering for the study of cell membrane transport

Objective

In this application we propose to apply the emerging tools of solution-phase, time-resolved X-ray scattering to the study of transfer phenomena in membrane transport proteins. Diffusive X-ray scattering of heavy metals in solution has recently been used by the host to monitor reaction kinetics of simple photochemical systems on a picosecond timescale. We seek to extend the technique to monitor structural dynamics inside the transport channel of halorhodopsin (hR), an important membrane protein that transports halides after photoactivation. The proposed experiment on hR will yield a movie of ion transport in the channel with the aim to clarify into the mechanism of trans-membrane ion transport. Success in the project will open new experimental windows for the study of basic cellular processes of tremendous scientific and biomedical importance.
