Final Report - MYCO-REG (Global regulation in Mycobacterium: Role of Lsr2)

This is the final report of the MY-COREG project, which is funded under the European Commission (EC) Seventh Framework Programme (FP7). The project focused on understanding the global regulation of mycobacteria, known to cause serious diseases in mammals, particularly on mutant gene Lsr2. MY-COREG combined transcriptome analysis, molecular genetic experiments and genomics to understand the global role of Lsr2. As Lsr2 negatively regulates the GlycoPeptidoLipid (GPL) expression level in mycobacterium smegmatis, beta-galactosidase fusions of full length and various deleted upstream promoter regions of GPL locus were constructed and the promoter activity in wild type and Lsr2 mutant strains were measured using a beta-galactosidase assay.

This project confirms the presence of GPLs in different mycobacterial species as an important component of virulence. The project has enhanced understanding of how GPLs are regulated, and its results also provide a very useful insight for other pathogenic species.
