Chromatographic behaviour of a peptide intrinsic to the metallothionein structure
The hexapeptide Lys-Cys-Thr-Cys-Cys-Ala [56-61] MT I (FT), which is a molecule intrinsic to the metallothionein structure, has been studied by reverse phase liquid chromatography, using electrochemical detection with a graphite electrode. After optimisation of the chromatographic separation, the behaviour of this compound containing three thiol groups has been investigated as a function of various parameters such as % of organic solvent, pH, time and temperature. The hydrodynamic voltammogram has been established. Due to the remarkable chemical properties of the SH group and the structure of the peptide, the addition of oxidising or reducing agents to the peptide solution before injection on a C18 column leads to new chromatograms. A mechanism of evolutive behaviour of this molecule is proposed.
