The certification of the catalytic concentration of lactate dehydrogenase isoenzyme 1 (LD1) (EC 126.96.36.199) in a reconstituted lyophilized material : CRM 404Funded under: FP3-MAT
The report describes the work carried out for the certification of the catalytic concentration of lactate dehydrogenase (LD) (L-lactate : NAD(+) oxidoreductase, EC 188.8.131.52) in a lyophylised solution of purified LD1 from human erythrocytes in a matrix consisting of human serum albumin and saccharose. This material can be used to verify the comparability of the results from laboratories using the SFBC method and for calibration and intra laboratory quality control of this measurement procedure. The preparation has been shown to be homogeneous, and no loss of activity upon storage at -20 C could be predicted on the basis of accelerated degradation studies. The catalytic concentration of LD in samples of the reconstituted material is certified to be (642 +/- 26) U/l or (10.7 +/- 0.5) micro-kat/l according to the recommended SFBC method at 30 C.
