Cytotoxicity and morphological transformation have been studied in BALB/3T3 cells for ammonium vanadate [vanadium(V)] and vanadyl sulphate [vanadium(IV)] alone or in combination with diethylmaleate (DEM), a cellular glutathione (GSH)-depleting agent. The materials and methods used are described under the following headings: (i) chemicals, radiochemicals and counting the radioactivity; (ii) cell cultures and media; (iii) cytotoxicity assay; (iv) transformation assay; (v) oxidation state of vanadium by electron paramagnetic resonance. The results are presented with details of the intracellular bioreduction of vanadium, cytotoxicity and morphological transformation.
