Chromatographic characterisation of a hexapeptide containing three thiol groups, intrinsic to the metallothionein structure
The hexapeptide lys-cys-thr-cys-cys-ala is a molecule intrinsic to the metallothionein structure. The chromatogram of the peptide consists of two peaks and this article presents the optimisation procedure for their separation. The relative proportion of the two peaks can vary with experimental conditions and is dependent on the peptide concentration. The effect of the proportion of organic modifier and the pH of the mobile phase led to the detection of the two species attributed to the two chemical forms of the peptide. It is believed that the hexapeptide is partially oxidised through an intramolecular disulphide bond.
