In this report further investigations in the handling of the thylakoid bioassay for herbicide determination in water samples are presented. Alterations on isolation and resuspending media led to higher activity due to a higher amount of scavenger. The measuring medium was concentrated in order to achieve maximum space for water samples, easy handling of the bioassay and clearer calculation of herbicide contamination. Evidently calculation of the mean underestimates detection possibilities of thylakoid suspensions. Measuring the yield 40 times directly after mixing the components and realization of integral calculation resulted in a better detection limit of 0.4 µg diuron per litre.
