Validation of analytical methods to determine the content of aflatoxin, ochratoxin and patulin in foodstuffs of vegetable originFunded under: FP4-SMT
An interlaboratory comparison was conducted to evaluate the effectiveness of an immunoaffinity column clean-up HPLC method for the determination of aflatoxin B1 and total afaltoxins at proposed European regulatory limits. The test portion of sample was extracted with methanol-water (8+2, v+v) for dried figs and paprika, and with methanol-water (8+2, v+v) plus hexane (or cyclohexane) for peanut butter and pistachios. The sample extract was filtered, diluted with water and applied to an immunoaffinity column. Samples, both naturally contaminated with aflatoxins and containing added aflatoxins, were sent to collaborators in 16 different European countries. Recoveries for total afloxins ranged from 71% to 92%, with the corresponding recoveries for aflatoxin B1 ranging from 82% to 109%. The method showed acceptable within-laboratory and between-laboratory precizions for all four matrices.
