The certification of the number of colony froming particles of Salmonella Typhymurium and number fraction of negative capsules from artificially contaminated milk powderFunded under: FP5-GROWTH
This report describes the certification of a second batch of the Salmonella typhimurium (ALM 40) reference material (CRM 507R). The number fraction of negative capsules and number content of colony forming particles of S. typhimurium from artificially contaminated milk powder were certified. Twelve laboratories participated in the certification study. Eleven laboratories determined the number of colony forming particles (CFP's) of Salmonella in a capsule. This was done according to a procedure developed for that purpose (enumeration procedure). In addition, the presence/absence of Salmonella CFP's in a capsule was determined by eleven laboratories, according to a procedure based on the ISO 6579 method. With the enumeration procedure the mean number of Salmonella CFP's and the fraction of capsules in which no Salmonella could be detected was determined. With the presence/absence procedure only the fraction of capsules in which no Salmonella could be detected. The certified contamination level is 5.0 Salmonella CFP in one capsule (two-sided 95% confidence interval 4.5 -5.4). The fraction of capsules in which no Salmonella could be detected, based on the enumeration procedure, is 1.1 % (one sided 95% confidence upper limit 0 -2.1 %) and 1.6 % (one sided 95% confidence upper limit 0 -2.8 %) based on the presence/absence procedure.
