Abstract

This report describes the production and certification of chlortetracycline (CTC)-free and CTC-incurred pig tissues (kidney, liver and muscle) as reference materials. High performance liquid chromatography (HPLC) with fluorescence detection of CTC and 4-epi-CTC was used for all tissue analyses. A preliminary study was performed to optimise the production control. In the main study, 500 vials each of CTC-free and CTCincurred liver (BCR-695 and �696), kidney (BCR-697 and �698) and muscle (BCR-706 and �707) were produced. A stability study was performed over 79 weeks testing samples stored at -18, +4, 20 and 37 °C. No measurable degradation occurred over this time period at any of the storage temperatures. Further, homogeneity of the materials was tested, too. Certified values of the �blank materials� were based on the observed detection limit. Certified values for the CTC-positive materials are accompanied by a GUMcompliant uncertainty statement (coverage factor k = 2) [1] considering uncertainty contributions from the batch characterisation, homogeneity, stability as well as from the common calibrant used in the certification exercise.
