Abstract

This report describes the preparation and testing of a reference column using two test procedures under high-performance liquid chromatography (HPLC) conditions. The reference column consists of silica xerogel beads with a particle diameter of (6 ± 0.5) µm which are chemically surface-modified with a C18 chain. The particles are packed into stainless steel columns of (150 x 4.0) mm dimensions.
The BCR-CRM is manufactured according to a standard operating procedure (SOP) developed in this project. The RM is in the form of one HPLC column and two test solutions A and B. The certification of the column is for its shape selectivity (chromatographic selectivity between o-terphenyl and triphenylene in test A), hydrophobicity (chromatographic selectivity between butylbenzene and pentylbenzene in test A) and ion-exchange activity (chromatographic selectivity between benzylamine and phenol in test B) using the supplied test solutions as shown below.
The certification was conducted between 8 independent laboratories. The method used was reversed-phase high-performance liquid chromatography (RP-HPLC) for the evaluation of the retention times, performance and selectivity of the analytes applied in the tests A and B. The certified uncertainty is expressed as the half-width of the 0.95 confidence interval.
