Certification of a Reference Material of Purified Genomic DNA from Escherichia Coli O157 (EDL 933), Certified Reference Material IRMM-449
This report describes the production and certification of a reference material of purified genomic DNA (gDNA) of Escherichia coli O157 strain EDL 933 (IRMM-449). This CRM supports the harmonisation and validation of PCR methods by their use as taxonomic controls in PCR reactions. The homogeneity and stability (at four different temperatures for up to 24 months) of the batch was assessed by monitoring amplification of the fliC gene by PCR, by agarose gel electrophoresis of gDNA and determination of the amount of gDNA per vial. The material is stable at �20 °C. The authenticity of the gDNA was confirmed by DNA sequence analysis of the fliC gene. Each vial contains an indicative mass of 1.3 µg freeze-dried gDNA, with an expanded uncertainty (UCRM) of 0.7 µg (k = 2).
