More detailed and more technical background information is given relating to this topic of the fifth call for proposals under the specific programme for standards, measurements and testing (SMT), 1994-1998. The reference method for the determination of the number of CFU, IDF-standard 100B/1991, has to be scrutinized, applying a ruggedness-test (slight deliberate variations of experimental conditions). As a result, a more precise method description will be available. A collaborative test of the improved method using skimmed milk powder samples is to be carried out. The data obtained allow the determination of the repeatability and the reproducibility limit, which are a prerequisite for the calculation of the "critical difference". The "critical difference" should be taken into consideration, when interpreting microbiological data. Expert statistical advice is needed for this and the following part of the project. The second part of the project aims at providing a reference material (milk powder) for the determination of the number of CFU. As a first step, conditions have to be found which allow to produce a material of sufficient homogeneity. On condition that this material is available, stability tests can be carried out. A time scale and expected result format are also described.
