Mid-Term Report Summary - EVOGENETICSTFBINDING (Evolutionary genetics of transcription factor binding in closely related mammals)

In the first half of the five years of this ERC Consolidator award, we have published over ten manuscripts that describe how genetics can direct transcription factor binding, and how different types of regulatory control interplay to generate a stable cellular phenotype. Most of these manuscripts arose from experiments focused on Aims 1 and 2. These results relied on the extensive use of what are known as reciprocal cross mice, where two strains are mated male to female versus female to male. Such an approach unlocked our ability to determine what influences were happening close to genes, as opposed to those that may be acting at a distance. Our work on Aim 3, wherein we will genetically alter the mouse genome using cutting-edge DNA modification tools to interrogate how very specific bases influence transcriptional regulation, has been focused on establishing these technologies, and so has not had any associated publications to date.
