Mid-Term Report Summary - DNAFOLDIMS (Advanced mass spectrometry approaches to reveal nucleic acid folding energy landscapes)

Our aim is to understand how nucleic acid sequences fold into tridimensional architectures, and how small molecules (cations or organic ligands) influence these pathways. To distinguish and characterize the different states present in complex mixtures, we develop approaches based on mass spectrometry. Using ion mobility spectrometry and molecular modeling, we could reveal how and why nucleic acid structures change when passing from the solution to the mass spectrometer. This work unveils fundamental principles of biomolecule mass spectrometry. DNAFOLDIMS also enabled us to construct a new instrument, coupling a wavelength-tunable laser with a mass spectrometer, to record electronic spectra on mass-selected ions. We also made significant progress in RNA analysis by mass spectrometry, in the presence of magnesium.
