| Abstract Mamba venoms have allowed the characterisation and purification of certain voltageactivated K+ channels from brain tissues and allowed their subunit and oligomeric structures to be described (see Proposal) A more detailed elucidation of the K+ charmed structure, including definition of secondary structure elements and components involved in K+ channel function (e.g. voltage sensing, inactivation and activation) will require the production of 2- and, in the long-term, 3-dimensional crystals. While using the purified, natural K+ channels may be desirable, producing enough material for such studies is a considerable undertaking. Thus, the aim of this proposed study is to systematically evaluate a number of systems for the overexpression of both alpha and beta subunits- singly and together- in an active, native form. When a suitable system has been found, 2-dimensional crystallisation trials will be performed, followed by electron microscopic analysis. Contract number : BIO4CT965117 | ||
