Structure-function relationships of the apoa-i protein: a multidisciplinary approach to the molecular basis of atherosclerosis

Objective



Project description of the work to be carried out in Pushchino (Russia): The proposed object of study is apolipoprotein A-I, its mutants and fragments as specified in the general description of the project. The Pushchino group, which has a long experience in conducting similar protein studies, would like to join the project and use the opportunity to work inside a team of leading European laboratories. The members of the Russian group are ready to cover the following aspects of the project. The thermodynamic parameters of protein folding and stability will be characterized by differential scanning calorimetry (DSC), isothermal titration calorimetry (IC) and by monitoring the temperature-induced changes of the optical parameters, such as circular dichroism and fluorescence. These studies will be carried out in tight collaboration with the Granada group, both in Granada and Pushchino, and will include the development of the DSC data analysis procedures and software by the members of the Russian team.
In addition the Pushchino group is planning to provide the facilities for expression and purification of the protein, its mutants and folded fragments to be used in thermodynamic studies, which ususally consume relatively large amounts of highly purified samples. The Pushchino group is also able to perform the kinetic studies of protein folding by denaturants using conventional stop-flow techniques. The group has four high-sensitivity scanning calorimeters of various types, an isothermal calorimeter and optical instruments. The large-scale protein purification facilities will be provided by the Institute of Protein Research to which the group belongs.
