PRODUCTION OF YEAST CAPABLE OF CONVERTING CRYSTALLINE CELLULOSE TO ETHANOL

Objetivo

FOR THE PAST NUMBER OF YEARS WE HAVE STUDIED A CELLULASE SYSTEM PRODUCED BY A FUNGUS, TALAROMYCES EMERSONII CBS 814.70. THIS CELLULASE SYSTEM IS CAPABLE OF CONVERTING CELLULOSE TO GLUCOSE AND THE RESULTING GLUCOSE MAY THEN BE CONVERTED TO ETHANOL USING YEAST. OUR PRIMARY OBJECTIVE IS TO INTRODUCE THE GENETIC INFORMATION CODING FOR THE CELLULASE COMPONENTS IN T.EMERSONII INTO ETHANOL PRODUCING YEAST STRAINS SUCH THAT THOSE STRAINS MAY THEN CARRY OUT THE CONVERSION OF CELLULOSE TO ETHANOL. IT IS ALSO AN OBJECTIVE TO INTRODUCE THE MOST EFFICIENT COMPONENTS INTRODUCED BY THE ORGANISMS C.THERMOCELLUM AND TRICHODERMA KONINGII INTO YEAST. THIS LATTER OBJECTIVE INVOLVES A COLLABORATION WITH DR G HALLIWELL, SWANSEA, WALES.

SINCE THE START OF THIS PROJECT IN DEC. 1986 WE HAVE ISOLATED SOME OF THE CELLULASE COMPONENTS PRODUCED BY T.EMERSONII. A NUMBER OF SPECIFIC MONOCLONAL ANTIBODIES HAVE BEEN PRODUCED TO THOSE COMPONENTS WHICH WE BELIEVE MAY FUNCTION AS PROBES FOR GENE BANKS IN ORDER TO ISOLATE THE GENES CODING FOR THOSE COMPONENTS. IN ADDITION WE HAVE DEVELOPED A ZYMOGRAM STAIN FOR EXOCELLOBIOHYDROLASE PRODUCED BY T.EMERSONII. THIS WORK WILL BE PRESENTED AT THE CONFERENCE ON ENERGY FROM BIOMASSIN ORLEANS, FRANCE IN MAY, 1987.
