Models of oxidative dna damage associated with active oxygen species


Research objectives and content
Active oxygen species play an important role in a large number of human conditions caused by oxygen toxicity. In this project, the question of whether superoxide anion, generated as a result of radical induced DNA damage involving the C-1' position, is capable of inducing further damage within the lipophilic environment of the DNA grooves will be examined. We will utilize synthetic modified nucleosides and oligonucleotides, able to generate superoxide anion, and we will study their reactions under physiological conditions.
Training content (objective, benefit and expected impact)
The training benefit of the previous category 30 grant can be transferred to the host institution and I will gain from the expertise of the host laboratory in the area of active oxygen species. The expected results will lead to a better understanding of the oxidative DNA damage mechanisms. The new modified nucleosides will benefit the chemistry of nucleosides and may provide new leads for the pharmaceutical industry.
