Development of novel electrochemical biosensors for glycoalkaloids determination in foodstuffs and biological samples

Objective

The development of reproducible, reliable, cheap and simple methods for monitoring of toxic compounds is of increasing importance to assure high quality and safety of foodstuffs and to reduce toxicity of different chemicals on living organisms. Routinely used methods for steroidal glycoalkaloids detection have a lot of disadvantages such as poor assay specification, their time-consuming nature and often are highly expensive. Biosensors seem to be a very promising tool to overcome the problems mentioned above. Moreover, the biosensors for steroidal glycoalkaloids detection are sill unavailable neither in the scientific laboratories nor at the market place. Therefore, this project is aimed to develop scientific and technological basis for creation of novel enzyme electrochemical microsensors with given characteristics for (-solanine, (-chaconine and solanidine determination. Special attention will also be paid to combine the main principles of bioselective membranes forming with those of microelectronic technology, to utilise cheap materials and wastes of microelectronic production, to simplify the procedure of sensors manufacturing.

To create biosensors the following materials and methods will be used: pH-sensitive field effect transistors, amperometric and conductometric interdigitated planar microelectrodes as transducers; acetylcholinesterase, butyrilcholinesterase, choline oxidase and peroxidase as bioselective materials; potentiometry, amperometry and conductometry as electrochemical registration methods. The study of biosensors developed will be performed in two steps: first through the investigation of measurement conditions using model solutions of the steroidal glycoalkaloids and then the examination of the sensor behaviour for the quantitative monitoring of such toxic compounds in foodstuffs and biological samples.

In order to achieve the main goal of the project, research activities will be focused on the following main research tasks:
- comparative analysis of different transducers for creation of glycoalkaloid specific biosensors;
- development of the methods of enzyme immobilisation and stabilisation on different transducer surfaces;
- development of laboratory prototypes of electrochemical sensors sensitive to (-solanine, (-chaconine and solanidine and optimisation of their performances;
- investigation of the enzyme-inhibitor interactions by means of standard and electrochemical methods;
- development and optimisation of measurement protocols for glycoalkaloids detection in foodstuffs;
- investigation of the biological action of various dozes of potato glycoalkaloids on organism of rats and possibility of the biosensors application for assessment of glycoalkaloids level in mammals organism.

The outcome of the research proposed is very important for both EU and NIS countries, since it will bring new, efficient and smart analytical devices in the market, will give the possibility to increase food quality and food safety, and will provide a good chance for future improvements in human and animal health.
