A Novel Olfactory Secretory Enzymes: Identification, Structure, and Function

Objective



Recently, novel specific water-soluble 45-kDa GTP-binding and 28-kDa thiol-specific antioxidant proteins were found in the rat olfactory epithelium by Fesenko's group. The goal of this project is to reveal the role of these secretory proteins in functioning of the receptor cell and their presumable function in the primary events in olfactory reception.

At the first stage, the project includes isolating these proteins, determining their structures, cloning of c-DNA that encodes these proteins, and homology searches through the databases that may allow to suggest ther presumable function. At the second stage, the following studies are planned:

comprehensive study of the enzymatic activity of these proteins (elucidation of substrates, co-factors etc.)
the localization of the 45-kDa and 28-kDa proteins in the olfactory epithelium at both the light microscopic and electron microscopic level
studies of metabolism of these proteins. The obtained data should allow to reveal the possible relationship between 45-kDa and 28-kDa proteins and other components involved in primary events of odor recognition in olfactory cell function.

Results obtained in this work will be published as scientific articles.
