THE CONTROL OF FOMES ANNOSUS AND THE INTERACTIONS OF THE FUNGUS WITH ITS HABITAT

Objectif



THE PROPOSAL FOR THE INVESTIGATION OF THIS DISEASE IS FIRSTLY TO CONTINUE THE DETAILED SEARCH FOR BIOLOGICALLY ACTIVE COMPOUNDS BY PHYTOCHEMICAL, EXAMINATION OF THE ORGANISM; SECONDLY TO STUDY THE ROLE OF THE ORGANISM'S INTERACTION WITH THE HOST, AND THIRDLY TO DESIGN A SELECTIVE FUNGICIDE WHICH IS STRUCTURALLY RELATED TO THE KNOWN HOST RESPONSE COMPOUNDS (PHYTOALEXINS). THE PRELIMINARY PHYTOCHEMICAL INVESTIGATION BY THIS RESEARCH GROUP ON ISOLATES OF FOMES ANNOSUS (OBTAINED IN IRELAND, THE NETHERLANDS, THE UNITED KINGDOM AND THE UNITED STATES OF AMERICA) HAS IDENTIFIED TWO METABOLITES WITH BIOLOGICAL ACTIVITIES;

(I) FOMANNOXIN PRODUCED BY THE ACTIVELY GROWING FUNGUS HAS A TOXICITY, TO THE UNICELLULAR ALGA CHLORELLA PYRENOIDOSA, TO GERMINATION SPRUCE SEEDS, AND TO YOUNG SPRUCE SEEDLINGS AND

(II) FOMAJORIN PRODUCED IN AGEING CULTURES AND WHICH SHOWED A MARKED ANTAGONISM TO OTHER FUNGI. THE BIOSYNTHESIS WILL BE INVESTIGATED, AND A HIGH YIELD SYNTHESIS OF BOTH THESE ACTIVE COMPOUNDS WILL BE DESIGNED. THE SYNTHETIC MATERIALS WILL BE USED FOR BIOLOGICAL STUDIES.

THE BIOSYNTHESIS STUDIES WILL ENTAIL THE FEEDING OF 14C AND 13C PRECURSORS.13C FT-NMR SPECTROSCOPY WILL BE APPLIED TO ASSIGN THE SYNTHESIS PATHWAY OF THE METABOLITES. THE CATABOLISM OF THE METABOLITES WILL BE TRACED BY REFEEDING THE LABELLED NATURAL PRODUCTS. THE VIVOTOXIN, FOMANNOXIN IS POSSIBLY DETOXIFIED BY HYDROXYLATION OF THE ISOPENTENYL SIDE CHAIN AND/OR BY OXIDATION OF THE AROMATIC ALDEHYDE GROUP. A SEARCH FOR THOSE OXIDATION PRODUCTS WILL BE CARRIED OUT USING AS MARKERS STANDARD SAMPLES FROM THE SYNTHETIC (+-) AND (+) FOMANNOXIN. THE BIOLOGICAL ACTIVITY OF THE OXIDATION PRODUCTS WILL BE EXAMINED. THE PROPOSED AND UNUSUAL BIOSYNTHETIC PATHWAY FOR THE NOVEL ISOCOUMARIN, FOMAJORIN S IS FROM MEVALONATE VIA A PROTOILLUDYL CATION OR ITS EQUIVALENT. THE OTHER NATURALLY OCCURRING ISOCOUMARINS (USUALLY FOUND IN THE FUNGI IMPERFECTI AND ACOMYCETES) ARE DERIVED FROM POLYKETIDE.
