Cel THE PROJECT WILL PURSUE THE BIOCHEMICAL ANALYSIS OF DNA REPAIR IN HUMAN AND MAMMALIAN CELLS, WITHIN A JOINT PROGRAMME . OUR CONTRIBUTION TO THIS COLLABORATION WILL BE FOCUSED ON THE INVESTIGATION INTO THE PRIMARY EFFECTS OF RADIATION, VIZ., THE REACTION PRODUCTS FORMED WITH THE INTRACELLULAR TARGET, THE DNA IN THE CHROMOSOMES. WE AIM AT THE DETECTION, IDENTIFICATION AND QUANTITATIVE DETERMINATION OF THESE PRODUCTS. Radiation induced lesions were detected using 2 approaches. Firstly, immunochemical methods were used, which included the production of antibodies against radiation induced lesions and the development of immunochemical assays based on the antisera were obtained. Secondly, biochemical methods were used, which included the development of an alkaline elution method for the sensitive detection of single strand breaks, using fluorometric quantisation of deoxyribonucleic acid (DNA). The methods developed were then used in studies of the induction and repair of DNA lesions in cells exposed to ionising radiation of UV, either in vitro or in vivo.Immunochemical and biochemical methods have been developed and applied in studies on the induction and repair of deoxyribonecleic acid (DNA) lesions in cells exposed to ionising radiation or ultraviolet (UV) radiation either in vitro or in vivo.
