Identification of candidate protective molecules of echinococcus granulosus and the development of combined salmonella vaccines

The aim of this joint research is to develop a vaccine for E.granulosus. Preleminary results obtained at Immunology Department in Uruguay suggest that some parasite molecules may be relevant in evasion of host immune response. Apart from some protective activity exhibited bY protoscolex surface antigens, two kinds of parasite molecules Were selected for further analysis: proteases and anti complementary (anti-C) molecules. Challenge experiments, both in intermediate and definitive host, are also under Way. For these reasons new research must be oriented towards the detection oE protectiVe molecules to be used both in intermediate and definitive host infections. Those molecules Will be selected according to previous results related to E. granulosus and other helminths. .
Those selected antigens that are glycoproteins Will be expressed using as a host Aspergillus nidulans in Dr. Scazzochio's laboratory that Was one of the first to develop transformation techniques in that fungus. Expression of recombinant antigens in Salmonella Vaccines is not straightforward and requires Eurther development. Techniques for optimization of expression of parasite antigens Will be pursued using both antigens currently being investigated in Dr. Hormaeche's laboratory from parasites such as S. mansoni, and also candidate hydatid immunogens. The aim is to obtain maximal expression of the recombinant antigens in the salmonella vaccine in an undegraded and maximally immunogenic form. optimal methods of expression have not Yet been determined, and various systems are available in Dr. Hormaeche's laboratory for expressing recombinant antigens intracellularly, in the periplasm, and in the flagella. The immunogenicity of the recombinant antigens expressed with these novel Systems Will be studied in the mouse model, both With respect to humoral and cellular immune response.
