Comparative study of chromosome aberrations (stable translocations) induced by genotoxic chemicals in mouse somatic and germ cells using chromosome specific libraries and fluorescent in situ hybridization (FISH)

Obiettivo

1) Generation of chromosome specific DNA libraries for mouse and evaluate chemically induced translocations by fluorescent in situ hybridization in somatic and germ cells.

2) To study the influence of telomeric repeat sequences on the formation of chromosomal aberrations in somatic and germ cells of mice.


Mouse chromosome specific DNA libraries will be generated by flow sorting specific chromosomes from animals carrying Robertsonian translocations, isolating DNA and amplifying that by linker-adapter PCR technique. These probes will be used to paint specific chromosomes by FISH technique. The frequencies of translocations involving these specific chromosomes in bone marrow as well as mouse spermatogonial cells following in vivo treatment with ENU, MMS, mitomycin C, cyclophosphamide, bleomycin and 6 mercaptopurine will be evaluated.

FISH technique with telomere specific repeats will be employed to check the influence of these repeats on formation of chemically induced chromosomal aberrations, especially in germ cells, where it is known that these sequences are amplified.
