Development of a high through-put(250-350)automated dna sequincing system and sequincing of human cDNA and polymorphic micro -satellite dna clones

Objective



The objective of this additional proposal is to characterize a portion of human chromosome 21 (21q22) using technology that is being developed in the project of the coordinator.
The additional proposal Will test the reliability and day-to-day usefulness of the newly developed technologies in characterizing human genome. Since We are interested in studying abnormal expression of human cystathionine beta-synthase (CBS) We propose to analyze region 21q22.3 containing this gene. In the study We Will employ the YAC and pl phage libra
ries to obtain clones positive for CBS. We will further characterize the gene bY restriction mapping' splicing-Alu PCR' splicing-exon PCR' and automated DNA sequencing. Additional studies Will include the search for microsatellites and polymorphic markers and also functional analysis of CBS MRNA isoforms.
Individual tasks of the project and data analysis Will be carried out in close collaboration among the proposing laboratory, coordinator and prof.Estivill's laboratory.
